An analysis of the adhesion-molecule requirements for the rolling, arrest and transmigration of resting CD4 cells on activated endothelial monolayers under flow conditions. • Entry of naive CD4 T cells into peripheral lymph nodes requires L-selectin
    • of outstanding interest. It is demonstrated in vivo that L-selectin is required for the entry of naive CD4 cells into peripheral lymph nodes and that other adhesion molecules cannot replace the function of L-selection in this regard. The results show that the naive population is involved in continuous recirculation, which is required not only for extravasation but also for the generation of a primary immune response.

    • Distinct roles of L-selectin and integrins α4β7 and LFA-1 in lymphocyte homing to Peyer's patch-HEV in situ: The multistep model confirmed and refined
    • An in vivo demonstration of the multistep model for lymphocyte extravasation. Video microscopy in PP in situ shows sequential, but overlapping, requirements for L-selectin and α4β7 in mediating rolling, and G-protein-linked activation of α4β7 and LFA-1 in mediating arrest. In addition, it is shown that α4β7 alone can mediate both rolling and sticking of activated lymphocytes, suggesting that ligand densities play a role in the tissue-specific migration of antigen-experienced cells.

    • α4 integrins mediate lymphocyte attachment and rolling under physiologic flow
    • of outstanding interest. It is shown that α4β7 mediates lymphocyte attachment and rolling under flow conditions in vitro via both MadCAM-1 and VCAM-1, whereas α4β1 interacts only with VCAM-1. The results indicate that selectins and α4 integrins can share overlapping functions. The β2 integrin LFA-1, however, is not capable of initiating lymphocyte contact, a conclusion reached by these authors in another study in vivo [13].

    • of special interest. A demonstration that CD40 ligand can upregulate CD40, induce E-selectin and ICAM-1, and with IL-1, induce VCAM-1 on endothelium. The study suggests that interactions between adhesion/accessory molecules expressed by activated T cells and their ligands on endothelium can stimulate vasculature to express receptors that facilitate recruitment of T cells from blood at sites of inflammation.

    • Bacterial superantigens induce T cell expression of the skin-selective homing receptor, the cutaneous lymphocyte-associated antigen, via stimulation of interleukin 12 production
    • of special interest. This intriguing study shows that induction of CLA, a ligand for E-selectin, on T cells activated by bacterial toxins (superantigens) is attributable to production of IL-12. The results suggest a direct link between infections and lymphocyte homing in superantigen-mediated skin diseases, by highlighting a potential mechanisms for recruitment of specific subsets of T cells.

    • Adhesion between epithelial cells and T lymphocytes mediated by E-cadherin and the alpha E beta 7 integrin
    • of outstanding interest. The αEβ7 integrin, which is expressed primarily on intraepithelial lymphocytes, is shown to mediate heterotypic adhesion with epithelial cells through interaction with E-cadherin. This study demonstrates a role for integrins in tissue-specific retention of lymphocytes, and highlights the important concept that whereas many lymphocytes are engaged in recirculation or migration to inflammatory sites, cells must also utilize mechanisms to remain in tissue during a response that is mediated by adhesion molecules.
