In virtually all cases we have examined, the mercaptopropanoyl-containing conformationally restricted inhibitors are significantly less potent as ACE inhibitors in vivo (AI pressor) than their mercaptoacetyl counterparts, despite similar in vitro potencies versus ACE. Numerous examples have been cited in this laboratory (see refs 3a,b,e). For this reason we have concentrated our efforts in this work on the mercaptoacetyl class of inhibitors.
