Proc Natl Acad Sci USA 1995, 92:4753-4757. Together with [2*], this paper shows that overexpression of the PARP DNA-binding domain leads to elevated chromosomal instability and increased rates of spontaneous, and DNA-damage induced, sister chromatid exchanges. PARP DNA-binding domain overexpression also causes hypersensitivity to alkylating agents and γ-irradiation.

Genes Dev 1995, 9:509-520. This paper establishes that cells homozygous for null mutations in PARP lack PARP activity and are deviod of detectable poly(ADP-ribose). Other than being susceptible to epidermal hyperplasia and the development of skin lesions, PARP-deficient mice are reported to be healthy and fertile. Moreover, they are not hypersensitive to UV irradiation or DNA alkylating agents. These findings would argue against a simple role for PARP in DNA repair or checkpoint control mechanisms and have lead to a dramatic reappraisal of PARP function in vivo.
