J Clin Invest 1995, 95:477-485. This study exemplifies the concept of islet cell destruction via an amnestic response against viral antigens expressed on the cell surface from a previous infection. The cell damage appears to be mediated by cytotoxic T cells.

Diabetes 1994, 43:1260-1266. This excellent study demonstrated that serum from mice infected with the diabetogenic virus CVB4 strain and humans with IDDM reacted with GAD65, coxsackievirus P2-C protein and GAD65 peptides. It provides excellent evidence for the role of molecular mimicry in the pathogenesis of viral-induced diabetes.

J Clin Invest 1994, 94:2125-2129. This study further clarifies the role of molecular mimicry by showing that peripheral blood mononuclear cells from patients with IDDM reacted to both GAD65 and coxsackie P2-C peptides, whereas cells from normal controls did not. The study also showed that peripheral blood mononuclear cell reactivity to non-specific proliferative stimuli were not different between normal controls and patients with IDDM. In addition, the study defined the sequences with significant amino acid homology.

This study showed that Y cell crossreactivrty between GAD65 and coxsackie P2-C was accomplished only in the context of a diabetes susceptibility allele. Thus, the study supports the concept that the induction of diabetes requires both genetic susceptibility and crossreactrve T cell responses.

This study of diabetes in mice, induced by CVB4 stain E2, nicely demonstrated the multifactorial process of beta cell destruction. Macrophages, T cells and cytokines were all shown to play a role. In addition, the study showed that persistence of viral RNA in the pancreas was associated with diabetes.
