Significantly, the cleft palate is not accompanied by craniofacial defects, emphasising that the TGF-β3-deficiency phenotype results from an intrinsic primary defect in palatal shelf fusion, rather than as a secondary consequence of craniofacial abnormalities.

Nat Genet 1995, 11:344-346. An interesting paper, describing an alternative approach to identifying genes involved in cleft palate. The authors elegantly test the hypothesis that deletion of Gabrb3 alone, and not another gene within the deletion interval, causes cleft palate. The cleft palate phenotype is rescued by introducing a Gabrb3 transgene into mice homozygous for the deletion.

Mice homozygous for the mutation show no detectable collagen XI in their cartilage as a result of a single nucleotide deletion in the coding region of the Col11a1 gene.
