    • Matrix metalloproteinases and their inhibition in periodontal treatment
    • Ryan ME, Ramamurthy NS, Golub LM: Matrix metalloproteinases and their inhibition in periodontal treatment. Curr Opin Periodont 1996, 3:85-96. This recent review presents all current work pertaining to the inhibition of MMPs as a therapeutic modality for periodontal disease. The structure, activation, and specificity of the family of MMPs and the regulation of their expression are discussed. The relationship between MMP activity and periodontal pathogenesis is summarized, including the sources of MMP production regulated by cytokines and growth factors and the temporal relationship of gingival MMP activity and alveolar bone loss. A section is dedicated to both endogenous and synthetic inhibitors. The multiple mechanisms of MMP inhibition by tetracycline analogues are detailed. Both animal models and human studies relevant to the proposed role of inhibitors in the treatment of periodontitis are presented. The numerous medical implications of the nonantimicrobial anti-MMP properties of tetracyclines are listed.
