2,3 Pyrrolidinedicarboxylates as Neurotransmitter Conformer Mimics: Enantioselective Synthesis via Chelate-Controlled Enolate Alkylation
-
(a) Humphrey, J. M.; Bridges, J. M.; Hart, J. A.; Chamberlin, A. R. 2,3 Pyrrolidinedicarboxylates as Neurotransmitter Conformer Mimics: Enantioselective Synthesis via Chelate-Controlled Enolate Alkylation. J. Org. Chem. 1994, 59, 2467-2472. We note that this study, describing an elegant and more efficient preparation of both 16 and 25, appeared just after the completion of our syntheses of these proline analogs. This alternate route is conceptually identical to the one described herein, employing a stereocontrolled β-alkylation of a protected aspartic acid derivative. The N-phenylfluorenyl protecting group was employed in the synthesis. The relative stereochemistry of the alkylated material was proved by X-ray crystallography. A direct comparison of our intermediates with those described in this reference was therefore precluded, although the spectroscopic and chromatographic properties of 16 and 25 are identical to those reported. We recommend this prior approach for the large-scale preparation of 16 and 25.

Recent isotope partitioning experiments (P.-H. Tari, unpublished results) suggest that ATP binds before aspartic acid. Therefore it is likely that the analogs are competing with aspartate for the E. ATP complex.
