In this paper, HL60 cytosol is shown to contain PLD activity that was stimulated by ARF but not RhoA and cdc42H. Membranes also contained a GTPγS-stimulated PLD activity that could be inhibited by removal of RhoA by Rho-GDI. In addition, the membrane activity could be partially recovered by readdition of RhoA, cdc42, and Rac1. ARF also stimulated the membrane PLD activity and the combination of RhoA and ARF showed a synergistic effect. The data suggest the existence of at least two different PLD isozymes in HL60 cells.

This paper describes the ability of RhoA and cdc42 to activate a partially purified PLD activity alone; in the additional presence of ARF, these Rho proteins are synergistic. In addition, a third cytosolic component is described that is not dependent on guanine nucleotides.

J Biol Chem 1995, 270:2431-2434. Proteins in both membranes and cytosol are needed to observe GTPγS-stimulated PLD activity. One cytosolic protein that has been identified is ARF. This paper reports on the presence of another cofactor, a 50-kD protein in leukocyte cytosol that also supported GTPγS-stimulated PLD activity in leukocyte membranes. The 50-kD factor was found to be synergistic with ARF in its ability to activate PLD. This paper should be read in conjunction with that by Bourgoin et al. (J Biol Chem 1995, 270:3172-3178).
