A patient (referred to as S.M.) with bilateral amygdala damage due to Urbach-Wiethe disease was asked to rate the intensity of an emotion expressed in pictures of faces. Impairments included an inability to judge the intensity of certain expressions (fear and surprise) and an overall pattern of endorsing only prototypical expressions; this patient did not perceive similarities between expressions that are normally endorsed for more than one emotional category by control subjects.

Patients with amygdala damage had a relative absence of the skin conductance conditioned response (SCR) to a CS (tone) that signaled the US (loud noise).

Science 1995, 269:1115-1118. This study tested classical conditioning of a skin conductance response in a several patients with differing localization of medial temporal lobe damage. Impaired conditioning was observed in a human subject, patient S.M., with bilateral damage of the amygdala; this subject failed to acquire conditioned SCRs but maintained the ability to report facts about the cues used in the conditioning task. In contrast to this pattern of results, a subject with restricted bilateral hippocampal damage exhibited normal autonomic conditioning but had little declarative knowledge about the events in the conditioning task. The findings were interpreted as support for the notion that the amygdala in humans is involved in implicit emotional learning but not explicit (declarative) memory.

Monkeys with either selective bilateral excitotoxic lesions or aspiration lesions of the anterior rhinal cortex, a region that is ordinarily damaged in the process of lesioning the amygdala by aspiration, were assessed on a delayed nonmatching-to-sample task (DNMTS) and a test of food preference. Monkeys with anterior rhinal cortex lesions displayed a visual memory impairment on the DNMTS task and normal food preference. Conversely, monkeys with amygdala lesions displayed abnormal patterns of food preference and intact visual memory performance.

An increase in the expression of the early-immediate gene, c-fos, was evident in a large number of widely dispersed cortical and subcortical structures following fear conditioning. Pretreatment with the anxiolytic drug, diazepam, dose-dependently reduced conditioned fear responding and increased fear-induced c-fos expression in the central nucleus of the amygdala. Conversely, pretreatment with diazepam produced dose-related decreases in conditioned fear-induced c-fos in the majority of other cortical and subcortical structures originally activated.

This observation supports the role of thalamic inputs to the amygdala as an important basis for conditioned neural activity.

Fear potentiation of startle was measured after Pavlovian pairing of a CS and aversive footshock. Both lesions produced comparable impairment in fear conditioning to both auditory and visual CSs.
