This study showed that GBR12909 antagonizes the rise in extracellular dopamine produced by systemic cocaine, suggesting that dopamine reuptake inhibitors may be useful pharmacological adjuncts in the treatment of cocaine addiction and withdrawal.

The authors indicated that the discriminative stimulus of 3 mg/kg of cocaine is centrally mediated and pharmacologically specific. There is an enhanced involvement of dopamine D-1 receptors in comparison with dopamine D-2 receptors. They also suggested the involvement of norepinephrine in the discriminative stimulus effects of low doses of cocaine.

Br J Pharmacol 1994, 113:1391-1399. Many of the behavioral effects of cocaine are thought to be mediated via dopamine re-uptake inhibition and a subsequent increase in dopamine activity. This study showed that some effects of dopamine uptake blockers on dopamine transmission, particularly those of nomifensine and cocaine, may be attributed to increased dopamine release.

In neuropathological specimens from victims of cocaine overdose compared with age-matched and drug-free control individuals, the most marked increase in cocaine congener binding was seen in the nucleus accumbens. Rosenthal plots of the saturation binding curves showed a significant increase in the apparent density of high-affinity sites on the dopamine transporter for the cocaine overdose individuals, with no change in the affinity or density of the low-affinity-binding component.

In this controlled clinical study, acute cocaine administration (30 mg intravenously) significantly increased ACTH mean peak amplitude, percent increase in peak amplitude, peak area, total peak area, and incremental peak height in eight men. The authors postulated that cocaine stimulates the release of corticotropin-releasing factor.

In this study, they concluded that clinically relevant concentrations of both anesthetics inhibit high-affinity 3H-CFT binding, and the data suggest overlapping sites for halothane and 3H-CFT.

This study investigated the effects of acute cocaine toxicity on systemic hemodynamics, myocardial metabolism, and regional organ blood flow in relation to the depth of isoflurane anesthesia in swine. Cerebral and spinal cord blood flows were depressed with cocaine administration, whereas other major organ blood flows were preserved with the exception of reduced splenic blood flow.

Hemodynamic responses to cocaine and two metabolites, cocaethylene and benzoylecgonine were studied in conscious ewes and fetuses, which were chronically instrumented to measure maternal and fetal aortic pressures, uterine artery blood flow and fetal cerebral blood flow. Cocaine and cocaine metabolites have important effects on maternal and fetal hemodynamics and fetal cerebral blood flow, which are not dependent on decreased uterine blood flow or fetal hypoxemia.
