Although most patients with lung cancer had detectable telomerase activity, some did not. This paper supports the idea that some cancers may be mortal tumors. First detection of telomerase activity using pleural effusions, indicating that simple diagnostic tests for telomerase (eg, bronchial or perhaps bladder washes) may be useful.

It also demonstrates that peripheral blood mononuclear cells (B and T cells) have low telomerase activity that increases with in vitro stimulation. First report that telomerase activity in normal peripheral blood mononuclear cells decreases with increasing age.

Science 1995, 269:1236-1241. Describes the cloning of the RNA component of human telomerase. The template region of human telomerase encompasses 11 nucleotides complementary to the human TTAGGG telomeric ends. Germline and tumor cell lines express more human telomerase RNA than normal somatic cells and tissues. Human cell lines that express a mutant form of telomerase generate the predicted mutant telomerase activity. HeLa cells transfected with an antisense human telomerase lose telomeric DNA and eventually begin to die.

    • Blasco MA, Funk W, Villeponteau B, Greider CW: Functional characterization and developmental regulation of mouse telomerase RNA. Science 1995, 269:1267-1270. Describes the cloning of the mouse telomerase RNA component. The template region of mouse telomerase RNA is shorter than the human telomerase template region. Telomerase RNA levels increase approximately 18-fold in the immortal cells derived from primary mouse fibroblasts. The RNA is detected in all newborn mouse tissues, but is downregulated during postnatal development. The presence of telomerase RNA correlates with the presence of telomerase activity in adult mouse tissues.
