The double mutant mice do not develop ulcerative colitis, showing the dependence on thymic-derived T cells for this disease. Additionally, cell transfer experiments show that the abnormal interleukin-2-deficient thymic-dependent cells (that cause disease) can be controlled by normal thymus-derived but not thymus-independent T cells in mixed chimeras.

Although Fas expression appears normal, the superantigen-stimulated cells do not apoptose in the presence of anti-Fas antibodies.

Eur J Immunol 1995, 25:3053-3059. The lymphoproliferative disorder in interleukin-2-deficient animals was investigated by inhibiting T cells in vivo with an antibody to the CD40 ligand. This treatment prevented the disease and indicated that inhibiting other stimulatory molecules on the T cell does not allow these hyperreactive cells to cause disease.

Blood and splenic T and B cells are markedly reduced, as are thymocytes. B lymphopoiesis is blocked at the transition of pro-B to pre-B cells in the bone marrow. In general, the remaining lymphocytes show normal cell surface phenotypes, indicating some level of interleukin-7-independent development. Additionally, as this mutation is not intrinsic to the lymphocytes themselves but to the microenvironment, the remaining lymphocytes are capable of stimulation by mitogens and cytokines.

Interleukin-7 receptor-deficient animals show many similarities to interleukin-7 ligand-deficient or γc receptor-deficient animals. However, some of the interleukin-7 receptor mutants show a more severely hypocellular thymus, indicating that other molecules interacting through the interleukin-7 receptor may be playing a role in development at stages earlier than interleukin-7. However, it is unclear why only some of the mutant animals exhibit this phenotype.
