T cells, but not B cells, are required for bowel inflammation in interleukin-2 deficient mice

Ma A, Datta M, Margosian E, Chen J, Horak I: T cells, but not B cells, are •• required for bowel inflammation in interleukin-2 deficient mice. J Exp Med 1995, 182:1567-1572. To investigate the cell types involved in intestinal inflammation and anemia in interleukin-2-deficient mice, Rag-2 mutant mice (lacking T and B cells) or JH-deficient mice (lacking B cells) were bred to the interleukin-2 mutant mice. interleukin-2 by Rag-2 mutant mice are disease free, and interleukin-2 by JH mutant mice develop ulcerative colitis but not anemia.

Immunopathology of interleukin (IL) 2- deficient mice: Thymus dependence and suppression by thymus-dependent cells with an intact IL-2 gene

J Exp Med 1995, 162:1769-1776. Interleukin-2-deficient mice were bred to athymic nu/nu mice. The double mutant mice do not develop ulcerative colitis, showing the dependence on thymic-derived T cells for this disease. Additionally, cell transfer experiments show that the abnormal interleukin-2-deficient thymic-dependent cells (that cause disease) can be controlled by normal thymus-derived but not thymus-independent T cells in mixed chimeras.

Normal clonal expansion but Impaired fas-mediated cell-death and energy induction in interleukin-2-deficient mice

Kneitz B, Herrmann T, Yonehara S, Schimpl A: Normal clonal expansion • but Impaired fas-mediated cell-death and energy induction in interleukin-2-deficient mice. Eur J Immunol 1995, 25:2572-2577. To investigate the basis for the intestinal immunopathology in interleukin-2-deficient animals, T cells were stimulated with superantigens. Activation proceeds normally, but deletion or the generation of anergy was impaired. Although Fas expression appears normal, the superantigen-stimulated cells do not apoptose in the presence of anti-Fas antibodies.

Generalized autoimmune disease in interleukin-2-deficient mice is triggered by an uncontrolled activation and proliferation in CD4 (+) T cells
-
Sadlack B, Lohler J, Schorle H, Klebb G, Haber H, Sickel E, Neolle RJ, • Horak I: Generalized autoimmune disease in interleukin-2-deficient mice is triggered by an uncontrolled activation and proliferation in CD4 (+) T cells. Eur J Immunol 1995, 25:3053-3059. The lymphoproliferative disorder in interleukin-2-deficient animals was investigated by inhibiting T cells in vivo with an antibody to the CD40 ligand. This treatment prevented the disease and indicated that inhibiting other stimulatory molecules on the T cell does not allow these hyperreactive cells to cause disease.

-
Vajdy M, Kosco-Vilbois MH, Kopf M, Kohler G, Lycke N: Impaired mucos•• al immune responses in interleukin-4 gene targeted mice. J Exp Med 1995, 181:41-54. Mucosal immunization, using a potent adjuvant, was diminished or absent in interleukin-4-deficient mice. Neither B cells nor T cells respond well to the mucosal immunization, but interestingly, both respond normally to systemic challenge. The poor mucosal response also correlated with an inability to form germinal centers in Peyer's patches.

Lymphopenia in interleukin (IL)-7 gene-deleted mice identifies IL-7 as a nonredundant cytokine
-
von Freeden-Jeffry U, Vieira P, Lucian LA, McNeil T, Burdach SE, Murray •• R: Lymphopenia in interleukin (IL)-7 gene-deleted mice identifies IL-7 as a nonredundant cytokine. J Exp Med 1995, 181:1519-1526. Defines interleukin-7 as a uniquely important cylokine for T- and B-cell development and expansion. Blood and splenic T and B cells are markedly reduced, as are thymocytes. B lymphopoiesis is blocked at the transition of pro-B to pre-B cells in the bone marrow. In general, the remaining lymphocytes show normal cell surface phenotypes, indicating some level of interleukin-7-independent development. Additionally, as this mutation is not intrinsic to the lymphocytes themselves but to the microenvironment, the remaining lymphocytes are capable of stimulation by mitogens and cytokines.

Defects in B lymphocyte maturation and T lymphocyte activation in mice lacking Jak3
-
Thomis DC, Gurniak CB, Tivol E Sharpe AH, Berg LU: Defects in B lym• phocyte maturation and T lymphocyte activation in mice lacking Jak3. Science 1995, 270:794-797. Molecules downstream of the γc receptor complex, such as the Jak family of kinases, are involved in transduction of signals into cells. Jak 3-deficient mice show abnormalities in lymphocyte development that indicate the importance of interleukin-7/γc/Jak 3 interaction.

Early lymphocyte expansion is severely impaired in interleukin-7 receptor-deficient mice
-
Peschon JJ, Morrissey PJ, Grabstein KH, Ramsdell FJ, Maraskovsky E, Gliniak BC, Park LS, Ziegler SF, Williams DE, Ware CB, Meyer JD, Davidson BL: Early lymphocyte expansion is severely impaired in interleukin-7 receptor-deficient mice. J Exp Med 1994, 180:1955-1960. The interleukin-7 receptor binds at least two ligands, interleukin-7 and TSLP. Interleukin-7 receptor-deficient animals show many similarities to interleukin-7 ligand-deficient or γc receptor-deficient animals. However, some of the interleukin-7 receptor mutants show a more severely hypocellular thymus, indicating that other molecules interacting through the interleukin-7 receptor may be playing a role in development at stages earlier than interleukin-7. However, it is unclear why only some of the mutant animals exhibit this phenotype.

-
Lin JX, Migone TS, Tsang M, Friedmann M, Weatherbee JA, Zhou L, • Yamauchi A, Bloom ET, Mietz J, John S, Leonard WJ: The role of shared receptor motifs and common Stat proteins in the generation of cytokine pleiotropy and redundancy by IL-2, IL-4, IL-7, IL-13, and IL-15. Immunity 1995, 2:331-339. Cytokine/receptor-induced signal transduction proceeds to activation of transcription factors, such as the STAT molecules. This study compares the similarities and differences of activation of the STAT family of transcnption factors by interleukin-2, interleukin-4, interleukin-7, interleukin-9, and interleukin-15.
