The authors show convincingly that a single nucleotide substitution in the binding site of the transcription factor Oct-1 on the LPL gene promoter almost totally abolishes the promoter activity and results, when present in a heterozygous form, in a clinical phenotype of familial combined hyperlipidemia.

Although not affecting the transcription rate, this variant results in diminished translation of the APO(a) mRNA into a mature protein.
