Proc Natl Acad Sci USA 1995, 92:4462-4466. The authors show convincingly that a single nucleotide substitution in the binding site of the transcription factor Oct-1 on the LPL gene promoter almost totally abolishes the promoter activity and results, when present in a heterozygous form, in a clinical phenotype of familial combined hyperlipidemia.

Arterioscler Thromb Vasc Biol 1995, 15:58-64. The nucleotide T instead of C is present at position +93 in the APO(a) gene promoter in approximately one out of seven individuals. Although not affecting the transcription rate, this variant results in diminished translation of the APO(a) mRNA into a mature protein.

Proc Natl Acad Sci USA 1995, 92:1851-1855. The 4G allele of the insertion/deletion polymorphism in the PAI(-1) promoter is associated with higher plasma PAI(-1) activity and myocardial infarction. The lower promoter activity of the 5G allele is proposed to be caused by its ability to bind a nuclear repressor protein.
