The full cDNA sequence of ERK, a new member of the receptor tyrosine kinase EPH family, is reported together with its chromosomal localization at 1p34-35 determined by fluorescent in situ hybridization. Expression of ERK mRNA was present in normal epithelial tissues and fetal, but not adult, brain. High levels of expression were observed in cell lines derived from epithelial cancers. A high percentage (75%) of gastric adenocarcinomas expressed high levels of ERK, with well-differentiated cancers expressing high levels more frequently than poorly differentiated.

Paraffin-embedded sections from 354 axillary lymph node-positive women treated with adjuvant chemotherapy for breast cancer (randomized trial 77-30-51 by Mayo-NCCTG) were semiquantitatively scored for c-erb B2 expression by immunohistochemistry. c-erb B2 expression was significantly correlated with degree of nodal involvement, higher nuclear grade, and estrogen receptor negativity. However, c-erb B2 was not found to be an independent predictor of disease-free or overall survival.

Proc Natl Acad Sci U S A 1994, 91:9141-9145. RAS requires farnesylation for its translocation from its cytosolic site of synthesis to the plasma membrane. This report is an example of the use of specific targeting of this oncogene activation to inhibit growth of transformed cells in vivo in a nude mouse model. Because this class of pharmaceutical is already safely being used in patients for the treatment of hyperlipidemias, these agents may rapidly move into treatment studies for RAS-dependent cancers in humans.
