    • Shah NM, Marchionni MA, Isaacs I, Stroobant P, Anderson DJ: Glial growth factor restricts mammalian neural crest stem cells to a glial fate. Cell 1994, 77:349-360. Addition of NDF (glial growth factor) to clonal cultures of rat neural crest cells grown in a complex medium reduces the number of clones containing neurons and glia, and increases the number of clones containing glia only. This study indicates that NDF acts as an instructive signal, making a neural fate less likely and a glial fate more likely.

Schwann cells cultured from human peripheral nerves proliferate in response to human NDF, and cAMP synergizes with NDF to promote Schwann cell proliferation and suppress fibroblast growth over several passages in culture. Thus, large numbers of cells can be obtained from human nerve biopsies (see also [15]).

    • Kioussi C, Gross MK, Gruss P: Pax3: a paired domain gene as a regulator in PNS myelination. Neuron 1995, 15:553-562. Pax-3 mRNA is present in non-myelin-forming postnatal mouse Schwann cells but not in cells that express the myelin protein MBP. Microinjection of Pax-3 expression vector results in downregulation of MBP. Elevation of cAMP, which stimulates myelin differentiation, suppresses endogenous Pax-3 expression. These experiments implicate Pax-3 as a negative regulator of myelination.

    • Zoidl G, Blass-Kampmann S, D'Urso D, Schmalenbach C, Müller HW: Retroviral-mediated gene transfer of the peripheral myelin protein PMP22 in Schwann cells: modulation of cell growth. EMBO J 1995, 14:1122-1128. This paper tests the theory that PMP22 might be involved in growth arrest by using retroviruses to overexpress PMP22 in Schwann cells and fibroblasts. In Schwann cells overexpressing PMP22, entry of quiescent cells into the cell cycle is significantly delayed, providing some evidence to support this theory.
