The trochlear nerve in vertebrate brain projects from ventral to dorsal, suggesting that trochlear axons might be guided by a repellent molecule secreted by the floor plate. This paper demonstrates that both the floor plate and netrin-1-expressing COS cells are able to repel trochlear axon growth cones in vitro. It will be interesting to know if these neurons express a vertebrate UNC-5 homologue.

Evidence is presented that the chemorepellent activity of the developing ventral spinal cord is specific for nerve growth factor (NGF)-responsive DRG sensory afferents, which project to the dorsal spinal cord, and has no affect on neurotrophin (NT)-3-responsive sensory afferents, which project to the ventral spinal cord. Semaphorin (Sema) III is shown to be expressed in the ventral spinal cord, and recombinant Sema III is shown to be a chemorepellant for NGF-responsive DRG afferents but not NT3-responsive sensory afferents. These results show that Sema III can function as a selective chemorepellant to direct the patterning of sensory afferents in developing spinal cord.

Using a PCR strategy, the genes encoding several murine proteins related to semaphorin III are identified and their embryonic expression analyzed. These semaphorins are differentially expressed in mesodermal and neuroectodermal tissues. Murine semaphorin D (a specific homologue of collapsin and human semaphorin III) is shown to inhibit sensory afferent axon extension.

This paper provides a detailed description of the spatial and temporal pattern of UNC-6 expression in C. elegans. The expression pattern in combination with the further analysis of unc-5, unc-6 and unc-40 mutant phenotypes suggests novel functions for unc-6 distinct from its ability to attract some and repel other axons that grow circumferentially on the body wall. These novel functions include an effect on axon decussation at the ventral midline and an effect on the ability of follower axons to grow along UNC-6-expressing pioneer axons (see text). This paper also provides the first structure/function analysis of the UNC-6 protein, showing, for example, that the EGF-like repeat related to domain V-2 of laminin is required for UNC-5-mediated dorsally oriented migrations.

These authors injected specific antibodies to Ng-CAM or axonin-1 (both expressed on commissural axons), or antibodies to Nr-CAM (expressed primarily by the floor plate) into the central canal of the chick spinal cord in ovo (stages 17 to 24). They then analyzed effects on commissural axon growth in stage 25 embryos (see Figure 2). Antibodies to Ng-CAM or axonin-1 altered commissural axon fasciculation, but the Ng-CAM antibodies did not cause axon pathfinding errors. Antibodies to Nr-CAM and to axonin-1 were unable to prevent commissural axon attraction toward the floor plate, but were able to prevent these axons from entering the floor plate region. These results indicate that an interaction between axonin-1 on commissural axons and Nr-CAM on the floor plate normally facilitates guidance of the commissural axons across the ventral midline.

Neuron 1996, 16:in press. This paper describes cloning of the commissureless gene and a cellular analysis of Comm protein expression during development of the Drosophila embryo. The results suggest a molecular mechanism for midline decussation by pioneer axons. Comm is a novel protein required for the formation of axon commissures in the CNS of Drosophila. In the wildtype animals, Comm is expressed by midline cells and taken up by axons as they cross the midline. In commissureless mutants, commissural axons begin to extend toward midline structures, then retract without crossing, suggesting that Comm is required for decussation. The ability of axons that cross the midline to take up Comm protein may be a common mechanism for decussation as a similar phenomenon has been reported in decussating commissural axons of the mouse spinal cord (see [23,24]).

This study also shows that in the absence of Fas III, muscle innervation is normal, suggesting that Fas III acts as a synaptic target recognition molecule for RP3 that can be compensated by other molecules.

This paper represents the first report of a molecular label on the tectum that is involved in axon guidance (by chemorepulsion of retinal axon growth cones) and that could be part of the guidance system proposed by Sperry, although the retinal-tectal matching mechanism presumably involves chemorepulsion rather than chemoattraction.

Neuron 1995, 14:973-981. The Eph-related receptor tyrosine kinase REK7 (the rat homologue of CEK7), is required for cortical neuron bundling in vitro. A REK7-lgG fusion protein is shown to block axon bundling in vitro and is used to purify a ligand, AL-1, for this receptor. Soluble forms of this ligand are also able to block axon bundling. These results strongly suggest that REK7 and AL-1 play an important role in axon fasciculation during the formation of neuronal pathways.
