Locus-specific amplification of HLA class I genes from genomic DNA: Locus-specific sequences in the first and third introns of HLA-A, -B and -C alleles
-
Cereb N, Maye P, Lee S, Kong Y, Yang SY: Locus-specific amplification of HLA class I genes from genomic DNA: locus-specific sequences in the first and third introns of HLA-A, -B and -C alleles. Tissue Antigens 1995, 45:1-11. This report describes locus-specific DNA sequences present in the first and third introns of the HLA-A, -B, and -C genes, thereby allowing locus-specific amplification. These data represent an important contribution for the development of HLA-A, -B, and -C molecular typing.

HLA-Cw allele analysis by PCR-restriction fragment length polymorphism: Study of known and additional alleles
-
Tatari Z, Fortier C, Bobrynina V, Loiseau P, Raffoux C, Charron D: HLA-Cw allele analysis by PCR-restriction fragment length polymorphism: study of known and additional alleles. Proc Natl Acad Sci U S A 1995, 92:8803-8807. Because of the imprecision of HLA-Cw serology, the biologic role of HLA-Cw in transplantation has been overlooked. A DNA-based HLA-Cw typing technique (PCR restriction fragment length polymorphism) was developed that allows the typing of over 22 different HLA-Cw alleles and identifies a series of new HLA-Cw alleles. The molecular identification of previously unrecognized HLA-Cw alleles and new HLA-Cw alleles (undetected by serology) has led to a reevaluation of the impact of Cw matching in transplantation.

Matching for MHC haplotypes results in improved survival following unrelated bone marrow transplantation
-
Tay GK, Witt CS, Christiansen FT, Charron D, Baker D, Herrmann R, Smith LK, Diepeveen D, Mallal S, McCluskey J, et al.: Matching for MHC haplotypes results in improved survival following unrelated bone marrow transplantation. Bone Marrow Transplant 1995, 15:381-385. A DNA-based HLA haplotype matching technique using non-HLA DNA polymorphisms in the MHC as markers of blocks of ancestral haplotypes identifies shared evolutionary related HLA haplotypes. Such haplotype matching appears beneficial in terms of lower incidence of GVHD and longer survival in a series of 45 BMT cases.

The probability of finding a suitable related donor for bone marrow transplantation in extended families
-
Schipper RF, D'Amaro J, Oudshoorn M: The probability of finding a suitable related donor for bone marrow transplantation in extended families. Stood 1996, 87:800-804. Based on the assumption that family donors are better donors because they share complete haplotypes with the recipient, this work suggests that a suitable donor should be sought among cousins and blood-related aunts and uncles (the extended family). This would reduce the search time. A computer program is proposed that calculates the probability of finding a donor in the extended family given the combination of haplotype frequencies and family composition.

N Engl J Med 1996, 286-291. The first report that a DNA polymorphism of the CD31 gene (coding for an adhesion molecule) that was assessed by direct DNA sequencing increases the risk of acute GVHD. The study was conducted retrospectively in 46 patients receiving bone marrow transplants from HLA-identical sibling donors.

Late infections after allogeneic bone marrow transplantation: Comparison of incidence in related and unrelated donor transplant recipients
-
Ochs L, Shu XO, Miller J, Enright H, Wagner J, Filipovich A, Miller W, Weisdorf D: Late infections after allogeneic bone marrow transplantation: comparison of incidence in related and unrelated donor transplant recipients. Blood 1995, 86:3979-3986. Late infections appear more frequent after allogeneic transplantation from unrelated donors (98 cases, 84.7%) than from HLA-identical siblings (151 cases, 68.2%). This may reflect the positive impact of HLA allelic and haplotype matching in controling the immune response against infectious agents.
