Tumor cell bystander killing in colonic carcinoma utilizing the Escherichia coli DeoD gene to generate toxic purines
-
Sorscher EJ, Peng S, Bebok Z, Allan PW, Bennett LL Jr, Parker WB: Tumor cell bystander killing in colonic carcinoma utilizing the Escherichia coli DeoD gene to generate toxic purines. Gene Ther 1994, 1:233-238. The bacterial DEOD gene was used to catalyze the conversion of a normally nontoxic adenine analogue. A bystander killing effect was reported in colon carcinoma cells expressing DeoD and treated with the 6-methylpurine substrate.

Regression of established breast carcinoma xenografts with antibody-directed enzyme prodrug therapy against c-erbB2 p185
-
Eccles SA, Court WJ, Box GA, Dean CJ, Melton RG, Springer CJ: Regression of established breast carcinoma xenografts with antibody-directed enzyme prodrug therapy against c-erbB2 p185. Cancer Res 1994, 54:5171-5177. The bacterial carboxypeptidase G2 enzyme was targeted to malignant breast cells overexpressing erbB-2 via a rat monoclonal antibody against the external domain of the human erbB-2 receptor. The authors used a mustard alkylating agent as a prodrug, and a dose-dependent tumor stasis or regression was reported.

Systemic gene therapy of murine melanoma using tissue specific expression of the HSVtk gene involves an immune component

Vile RG, Nelson JA, Castleden S, Chong H, Hart IR: Systemic gene therapy of murine melanoma using tissue specific expression of the HSVtk gene involves an immune component. Cancer Res 1994, 54:6228-6234. Cancer Res 1994, 54:5408-5413. Enhanced sensitivity of breast cancer cells to ganciclovir was achieved via transcriptional regulation of HSVTK expression with an enhancer sequence from the breast specific DF3 promoter. Evidence for a bystander effect was also shown.

Proc Natl Acad Sci U S A 1995, 92:2889-2893. This paper describes an in vivo cancer gene therapy approach to introduce cytokine genes into tumor-bearing animals by Accell (Agracetus) particle-mediated gene transfer.
