Mutation screening identified several heterozygous mutations of the SRY-related gene, SOX9, in both sex-reversed and non-sex-reversed CD individuals. This is evidence that CD s an autosomal dominant haplo-insufficiency syndrome and underlines the importance of gene dosage in sex-determining mechanisms.

This mouse knockout experiment of Sf1, known to be required for steroid hormone biosynthesis, demonstrates that the gene is also required for the development and maintenance of both the adrenal glands and the indifferent gonad. Mice deficient for Sf1 lack adrenal glands and gonads. The dual role of Sf1 in early gonadal formation and later in hormone production was unexpected.

Nature 1994, 372:635-641. This paper describes the cloning of DAX1 from the DSS-AHC critical region on the X. Mutations of DAX1 in AHC patients prove its identity as the adrenal hypoplasia congenita gene. DAXI is also a candidate for the X-linked dosage-sensitive sex-reversal gene and would encode a second nuclear hormone receptor (SF1 being the other) to be implicated in sex determination.

This paper describes the cloning and expression anajysis of mouse Dax1 In addition to transcription in the developing adrenal glands and hypothalamus, sexually dimorphic expression is seen in the gonad. Dax1 is down-regulated with overt differentiation of the testis in males, but persists in the developing ovary, providing circumstantial evidence for Dax1 being equivalent to DSS.
