Nucleotide binding to the hydrophilic C terminal domain of the transporter associated with antigen processing (TAP)
-
Muller K, Ebensperger C, Tampe R: Nucleotide binding to the hydrophilic C terminal domain of the transporter associated with antigen processing (TAP). J Biol Chem 1994, 19:14032-14037. The purified recombinant nucleotide-binding domain of TAP1 specifically binds to ATP and ATP analogues. However, ATP hydrolysis by TAP1 nucleotide-binding domain is not detected. These experiments support the theory of TAP-dependent peptide translocation.

-
Van Endert PM, Tampe R, Meyer TH, Tisch R, Bach J-F, McDevitt HO: A sequential model for peptide binding and transport by the transporters associated with antigen processing. Immunity 1994, 1:491-500. Characterization of TAP overexpression system in Sf9 cells demonstrates that TAPs are able to bind peptides in the absence of ATP and only when both TAP1 and TAP2 are present. Binding of ATP releases peptide from TAP peptide-binding site. Release of peptides from microsomes is ATP, pH, and temperature dependent.

Comparison of cell lines deficient in antigen processing reveals a functional role for TAP-1 alone in antigen processing

J Exp Med 1994, 180:1415-1425. CMT.64, a small-lung carcinoma cell line, does not express TAP1/2 unless treated with gamma interferon. Uninduced CMT.64, infected with vesicular stomatitis virus is not recognized by specific cytotoxic T lymphocytes. Introduction of rat TAP1 gene in CMT.64 restores cytotoxic T-lymphocyte recognition of infected cells, indicating possible involvement of TAP1 homodimer in peptide translocation.

Analysis of the fine specificity of rat, mouse and human TAP peptide transporters
-
Neefjes JJ, Gottfried E, Roelse J, Gromme M, Obst R, Hämmerling GJ, Momburg F: Analysis of the fine specificity of rat, mouse and human TAP peptide transporters. Eur J Immunol 1995, 25:1133-1136. Along with the study by Momburg et al. (Nature 1994, 367:648-651), this paper analyzes the effect of amino acid variation at the N- and C-termini and positions 2 to 7 of substrate peptides on TAP-mediated transport in streptolysin O-permeabilized cells and in microsomes. Only a few amino acids in internal positions significantly influence peptide transport. The C-terminal residue is the predominant determinant of species and substrate specificity.

Peptide size selection by the major histocompatibility complex-encoded peptide transporter
-
Momburg F, Roelse J, Hämmerling GJ, Neefjes JJ: Peptide size selection by the major histocompatibility complex-encoded peptide transporter. J Exp Med 1994, 179:1613-1623. As determined by Edman degradation of translocated peptides, TAP selects for peptides with the size range that covers the length of peptides associated with class I molecules. Longer peptides may also be transported, but some require prior trimming in cytosol.

Peptides that fail to bind to class I molecules are slowly trimmed in endoplasmic reticulum. Peptides released from the endoplasmic reticulum are rapidly degraded in the cytosol. A fraction of peptides that escape degradation recycle back to the endoplasmic reticulum in a TAP-dependent fashion.

Interaction of MHC class I molecules with the transporter associated with antigen processing
-
2-microglobulin complexes could be coimmunoprecipitated, using anti-TAP or anti-class I antibodies. TAP/class I association could be disrupted by addition of class I binding peptides. These data suggest that association of TAP with class I molecules may promote an efficient peptide capturing by the latter.

Herpes simplex virus turns off the TAP to evade host immunity
-
Hill A, Judovic P, York I, Russ G, Bennink J, Yewdell J, Ploegh H, Johnson D: Herpes simplex virus turns off the TAP to evade host immunity. Nature 1995, 375:411-415. Along with the paper by Fruh et al. (Nature 1995, 375:415-418), this paper provides evidence that cytoplasmic protein ICP47 of herpes simplex virus interferes with loading of class I molecules by direct binding to TAP and inhibition of peptide translocation.
