This study demonstrates that replacement of up to 50% of the apoA-I in HDL by the acute-phase reactant SAA, has no apparent effect on the ability of the HDL to promote cell cholesterol efflux. When virtually all of the HDL apoA-I has been replaced by SAA, the ability of the resulting HDL to promote cholesterol efflux is reduced by about 30%.

Arterioscler Thromb Vasc Biol 1995, 15:956-962. This study of A-I HDL demonstrates major differences between large and small particles in their ability to promote cell cholesterol efflux. Whereas small and medium sized A-I HDL are effective cholesterol acceptors, larger particles are not. In fact, the larger A-I HDL actually inhibit the efflux promoted by smaller particles.

    • Czarnecka H, Yokoyama S: Lecithin:cholesterol acyltransferase reaction on cellular lipid released by free apolipoprotein-mediated efflux. Biochemistry 1995, 34:4385-4392. Incubation of lipid-free apoA-I and apoA-II with macrophages and fibroblasts generated pre-β HDL. Although the newly formed A-I HDL were reactive with LCAT, the presence of LCAT did not enhance the efflux of cell cholesterol into the apoA-I-containing particles. LCAT did, however, inhibit the influx of cholesterol from HDL to cells, thus accounting for the ability of the enzyme to enhance the net efflux of cell cholesterol.
