J Exp Med 1995, 181:181-192. Linker-scanning analysis is used to identify an IL-4-responsive region in the germline ε promoter. A 27 bp DNA segment which binds IL-4 Stat/Stat 6 and a C/EBP transcription factor are found to be sufficient to transfer IL-4 inducibility to a heterologous promoter. It was also found that mutation of a NF-κB/p50 binding site reduces 40-fold the expression of the promoter, consistent with the lack of switching to IgE in p50 knockout mice.
