    • McGowan CH, Russell P: Cell cycle regulation of human WEE1. EMBO J 1995, 14:2166-2175. The molecular weight of human WEE1 is demonstrated to be approximately 94 kDa, which is substantially larger than the molecular weight of 49 kDa which was reported for the first human wee1 gene product to be identified. The authors show that WEE1 activity is strongly suppressed during M phase by a mechanism that requires phosphorylation of WEE1. Subcellular localization of p94 WEE1 shows that the protein is nuclear in location.

Antibodies generated against bacterially produced p49 WEE1 Hu recognized a protein of 95 KDa in HeLa cells, but did not recognize a 49kDa protein. The authors show that the 95 kDa protein phosphorylates Cdc2 on tyrosine 15, and that this WEE1 kinase activity is impaired during mitosis.

This report shows that overexpression of a kinase-defective c-Abl in fibroblasts both shortens the period of cell-cycle re-entry that follows the addition of serum to quiescent cells, and enhances transformation by Myc, Ras, v-Abl and v-Fms. The authors conclude that wild-type c-Abl is a negative regulator of cell growth.

An extension of the data presented in [26]. A mutant c-Abl that cannot bind pRb was created; this mutant is not cell cycle regulated. The activity of this pRb-resistant c-Abl is significantly elevated in quiescent cells, and remains high in S phase. In addition, the authors report that a positive growth effect is exerted by expression of wild-type c-Abl in human osteosarcoma cells. It is shown that overexpression of wild-type c-Abl can overcome pRb-induced growth arrest in these cells.
