The Ikaros gene is required for the development of all lymphoid lineages

Georgopoulos K, Bigby M, Wang J-H, Molnar A, Wu P, Winandy S, Sharpe A: The Ikaros gene is required for the development of all lymphoid lineages. Cell 1994, 79:143-156. Gene-targetting experiments identify a transcription factor, Ikaros, which acts as a master switch in haematopoiesis, directing cells along the lymphoid lineage. This discovery opens the way to the identification of the signal-transduction pathway that regulates Ikaros and thus controls lymphopoiesis.

Nature 1995, 376:435-438. The ZAP-70 protein tyrosine kinase is shown in knock-out mice to be essential for the transduction of TCR-mediated signals that lead to both positive and negative selection. These results complement studies of human SCID patients with a disrupted ZAP-70 gene [53,54], although the two phenotypes exhibit interesting differences.
