These workers showed that it was possible to colonize mice with H. pylori. The development of a murine model of H. pylori infection is an important contribution to vaccine development because it means that H. pylori-specific antigens will be able to be tested directly in mice.

Gastroenterology 1995, 109:115-121. In this study, mice that were already infected with H. felis were immunized with H. pylori urease B subunit, together with CT. Mice treated in this fashion were not only cleared of the infection, but also remained immune to subsequent challenges with H. felis bacteria. Therapeutic immunization was shown to be a feasible approach to the treatment of H. pylori gastritis.

J Infect Dis 1995, 172:161-172. This is a comprehensive study of the use of urease as a mucosal antigen to prevent H. felis infection in mice. Protection scores were compared with the humoral immune responses in the animals. It was shown that oral immunization was just as effective as the intragastric route in inducing protection, and also that very low doses of urease antigen were required to induce protection against H. felis infection.

This paper described a phase I clinical trial in which the safety of the antigen preparation (recombinant urease) was tested in patients who already had H. pylori disease. Urease was well tolerated by the patients, suggesting that this antigen represents a good candidate for a first generation vaccine.

In this study, incubation of an H. felis inoculum with monoclonal antibodies directed against H. felis antigens appeared to neutralize the ability of the bacteria to colonize mice. Several of the antibodies were shown to recognize epitopes of H. felis urease. Monoclonal antibodies of both IgA and IgG classes were able to neutralize H. felis bacteria.
