This paper details a set of experiments which use the AMPPNP-driven decatenation of a pair of singly interlocked duplex DNA rings to dissect the strand-passage reaction of a type II topoisomerase. The results provide strong evidence that the passed DNA duplex enters and exits the enzyme at different sites.
