The authors investigated the profiles of cytokine mRNA expression in inflammatory myopathy and concluded that sustained secretion of cytokines by T-cells or of IL-1 by macrophages is not a prerequisite for operation of the immune effector response and that muscle may not be the site of ongoing sensitization.

Clin Exp Immunol 1995, 100:519-528. PCR technique evaluated the γ/δ TCR gene expression in 45 muscle biopsies obtained from 42 inflammatory myopathy patients (17 polymyositis, 12 dermatomyositis, and 13 IBM), γ/δ TCR gene expression was noted in nine patients. However, no amino acid sequence similarities were evident within the V δ-N-D δ-N-J δ region of V δ I transcripts amplified from four patients suggesting that γ/δ T-cells rarely play a cytotoxic role in inflammatory myopathy.

A combination of PCR and double-fluorescence immunocytochemistry were used to analyse the TCR repertoire expressed among autoinvasive and nonautoinvasive T-cells in polymyositis patients. The results suggest that a strikingly limited TCR repertoire is expressed in polymyositic muscle; with dissociation between the TCR usage of autoinvasive and interstitial T-cells with clonal expansion of autoinvasive T-cells.

Arthritis Rheum 1995, 38: 690-698. All studied muscle biopsy specimen contained HTLV-I DNA. Two out of six samples demonstrated intense staining for HTLV-I gp46 in many of the invading mononuclear cells and weak staining for HTLV-I gp46 in many of the invading mononuclear cells and weak staining in the adjacent myocytes. Two other specimens were weakly positive for gp46 in rate mononuclear cells. All control specimens were negative for the presence of HTLV-I DNA and protein.

Pulmonary capillaritis, a distinct interstitial reaction associated with diffuse alveolar hemorrhage, occurs with the systemic vasculitides and some collagen vascular diseases. It has not been previously described in patients with polymyositis. Polymyositis and the pulmonary disease developed concomitantly with anti-Jo-1 antibodies.

Immunoscreening of the cDNA expression library, and cDNA isolation, alignment, and sequence analysis by Northern blotting and in-situ hybridization techniques were carried. A major antigen recognized by anti-Mi-2 positive sera of dermatomyositis patients was found to constitute a 218kd nuclear protein (218kd Mi-2) encoded on chromosome 12 and to belong to the helicase family.
