PCR technique evaluated the γ/δ TCR gene expression in 45 muscle biopsies obtained from 42 inflammatory myopathy patients (17 polymyositis, 12 dermatomyositis, and 13 IBM), γ/δ TCR gene expression was noted in nine patients. However, no amino acid sequence similarities were evident within the V δ-N-D δ-N-J δ region of V δ I transcripts amplified from four patients suggesting that γ/δ T-cells rarely play a cytotoxic role in inflammatory myopathy.

A combination of PCR and double-fluorescence immunocytochemistry were used to analyse the TCR repertoire expressed among autoinvasive and nonautoinvasive T-cells in polymyositis patients. The results suggest that a strikingly limited TCR repertoire is expressed in polymyositic muscle; with dissociation between the TCR usage of autoinvasive and interstitial T-cells with clonal expansion of autoinvasive T-cells.

Two out of six samples demonstrated intense staining for HTLV-I gp46 in many of the invading mononuclear cells and weak staining for HTLV-I gp46 in many of the invading mononuclear cells and weak staining in the adjacent myocytes. Two other specimens were weakly positive for gp46 in rate mononuclear cells. All control specimens were negative for the presence of HTLV-I DNA and protein.
