Blood 1995, 85:902-911. A new splice variant of G-CSF-R that is expressed at low levels in normal granulocytes is described. This variant is hampered in its proliferation and maturation signaling capacity. In one case of acute myeloid leukemia, this splice variant was overexpressed because of the presence of a point mutation in the cryptic splice donor site involved in the alternative splicing.

Studies with tyrosine substitution and C-terminal deletion mutants of G-CSF-R demonstrate that activation of STAT3, but not STAT1, requires the membrane-distal cytoplasmic region of G-CSF-R. Tyrosine 704 of G-CSF-R, present in a YXXQ consensus STAT3 binding sequence, is indeed involved in recruitment of STAT3. However, other domains in G-CSF-R that do not fit this consensus are also involved in STAT3 activation.

These nonsense mutations truncate the C-terminal domain essential for maturation induction. Because both normal and mutated alleles are expressed in leukemic cells, coexpression studies were performed in maturation-competent murine 32D cells, which demonstrated that the truncated receptor forms interfere with maturation signaling from the full-length (wild-type) G-CSF-R in a dominant negative fashion.
