    • Kennedy BK, Austriaco NR, Zhang J, Guarente L: Mutation in the silencing gene SIR4 can delay aging in S. cerevisiae. Cell 1995, 80:485-496. This study uses stress to screen for enhanced lifespan mutations and is analogous to earlier work with Drosophila. Stress resistance may be one of the few important unifying mechanisms that determine aging patterns. This is one of the few genetic studies of aging that makes some progress in addressing the mechanistic basis of postponed aging at the molecular level.

Nature 1995, 375:754-760. Cloning has supported the involvement of candidate gene AD3 in early onset (30-60 years) cases, where the gene has been mapped to chromosome 14q24.3. This paper shows that transcript S182 from this region is the most likely candidate gene, because its missense mutations co-segregate with AD.
