Association of a cytokine gene variant with an infectious disease; in a case-control study of Gambian children, homozygotes for a promoter variant at position -308 of the TNF gene were at a substantially increased risk of cerebral malaria. Asymptomatic carriers of hepatitis B virus in the Gambia were less likely to have the HLA-DRB1*1302 allele, suggesting that an immune response restricted by this HLA type may facilitate viral clearance. Recent analysis by Davenport et al. [51] of the sequence of peplides bound by this molecule should facilitate molecular immunological analysis of this HLA-disease association.
