The subcellular localization of defensins and myeloperoxidase in human neutrophils: Immunocytochemical evidence for azurophil granule heterogeneity
-
Oren A, Taylor JMG: The subcellular localization of defensins and myeloperoxidase in human neutrophils: immunocytochemical evidence for azurophil granule heterogeneity. J Lab Clin Med 1995, 125:340-347. In granules that contained either myeloperoxidase or defensins, colocalization was found in only one third. Another third contained defensins without myeloperoxidase and the rest contained myeloperoxidase without defensins. This finding establishes a link between functional and structural heterogeneity of azurophil granules.

Localization of human neutrophil peptide (HNP) and its messenger RNA in neutrophil series
-
Date Y, Nakazato M, Shiomi K, Toshimori H, Kangawa K, Matsuo H, Matsukura S: Localization of human neutrophil peptide (HNP) and its messenger RNA in neutrophil series. Ann Hematol 1994, 69:73-77. The amount of defensins is found to increase as neutrophils mature from promyelocytes to metamyelocytes. This indicates that there is a difference in timing of formation of the different azurophil granule subsets, established by Oren and Taylor (J Lab Clin Med 1995, 125:340-347.)

+ cells show a maturation-dependent difference in expression of messenger RNA for selected azurophil and specific granule proteins. The difference in appearance of messenger RNA for myeloperoxidase (early), elastase (intermediate), and defensins (late) is consistent with the heterogeneity of azurophil granules. Messenger RNA for specific granule proteins appears even later.

Blood 1995, 85:812-817. Normal bone marrow cells were separated according to their stage of maturation. Gelatinase was shown to be synthesized in more mature cells than lactoferrin and NGAL. Wortmannin binds specifically to 1-phosphatidylinositol 3-kinase while inhibiting guanine nucleotide-binding protein-coupled receptor signaling in neutrophil leukocytes
-
Thelen M, Wymann MP, Langen H: Wortmannin binds specifically to 1-phosphatidylinositol 3-kinase while inhibiting guanine nucleotide-binding protein-coupled receptor signaling in neutrophil leukocytes. Proc Natl Acad Sci U S A 1994, 91:4960-4964. Wortmannin, which inhibits signals generated through growth factor receptors and G-protein-coupled receptors, was shown to bind to the catalytic subunit of PI 3-kinase. In combination with the study by Ding et al. (J Biol Chem 1995, 270:11684-11691) and Stauanov et al. (Science 1995, 269:690-693), this article establishes a central role of PI 3-kinase in intracellular signaling in activated neutrophils.

Antagonists of phosphatidylinositol 3-kinase block activation of several novel protein kinases in neutrophils

Ding J, Vlahos CJ, Liu R, Brown RF, Badway JA: Antagonists of phosphatidylinositol 3-kinase block activation of several novel protein kinases in neutrophils. J Biol Chem 1995, 270:11684-11691. Several phosphatidylinositol 3-kinase inhibitors including wortmannin were shown to inhibit activation of several serine/threonine kinases and respiratory burst activation induced by chemoattractant receptor activation. This paper demonstrates the central role of phosphatidylinositol 3-kinase in neutrophil activation.

Cloning and characterization of a G protein-activated human phosphoinositide-3 kinase

Science 1995, 269:690-693. A phosphatidylinositol 3-kinase directly activated by heterotrimeric G proteins was cloned from a human bone marrow complementary DNA library. This may be the direct link between chemoattractant receptor stimulation and phosphatidylinositol 3-kinase activation, in particular if it can be shown that wortmannin also inhibits this phosphatidylinositol 3-kinase.

The exocytotic fusion pore of small granules has a conductance similar to an ion channel
-
Lollike K, Borregaard N, Lindau M: The exocytotic fusion pore of small granules has a conductance similar to an ion channel. J Cell Biol 1995, 129:99-104. Application of membrane capacitance studies to the cell-attached mode of patch-clamping allowed the resolution of distinct exocytotic steps corresponding to all known mobilizable organelles of neutrophils. Fusion pores of sizes similar to those of ion channels were demonstrated for all types of granules, indicating that proteins constitute the fusion pore in all granules and secretory vesicles.

The necrotic venom of the brown recluse spider induces dysregulated endothelial cell-dependent neutrophil activation-differential induction of GM-CSF, IL-8, and E-selectin expression
-
Patel KD, Modur V, Zimmerman GA, Prescott SM, McIntyre TM: The necrotic venom of the brown recluse spider induces dysregulated endothelial cell-dependent neutrophil activation-differential induction of GM-CSF, IL-8, and E-selectin expression. J Clin Invest 1994, 94:631-642. Spider venom gives a unique upregulation of endothelial cell proteins that induces adherence and activation of neutrophils but no transendothelial migration. This illustrates the significance of coordinating the different steps in neutrophil activation and provides and important tool for studying neutrophil-endothelial cell interactions.

The pro region of human neutrophil defensin contains a motif that is essential for normal subcellular sorting
-
Liu L, Ganz T: The pro region of human neutrophil defensin contains a motif that is essential for normal subcellular sorting. Blood 1995, 85:1095-1103. This study validates the use of the 32-Da c13 as a cell line for transfection of genes for granule proteins. It is demonstrated that the pro region of defensins is essential for sorting into a granule-like storage organelle.
