Autologous vascular smooth muscle cells were transduced with a retroviral vector containing rat EPO complementary DNA sequences. Rats transplanted with the transduced cells developed significantly increased erythrocytic indices that were sustained for over 2 months.

The nonparenchymal cells were identified as Ito cells, which were present in perisinusoidal sites with the space of Disse and showed structural similarities to the peritubular interstitial fibroblasts that produce erythropoietin in the kidney.

Pretreatment of hypoxic cells with hydrogen peroxide blocked induction of HIF-1 protein expression. Treatment of purified HIF-1 with oxidizing agents in vitro resulted in loss of DNA-binding activity. Redox state may therefore affect both hypoxia signal transduction and HIF-1 activity directly.
