    • Coleman RA, Taggart AK, Benjamin LR, Pugh BF: Dimerization of the TATA binding protein. J Biol Chem 1995, 270:13842-13849. Demonstrates that TBP forms dimers in a manner consistent with the crystal structure of TBP which is not bound to DNA. Importantly, dimerization is competitive with the ability of TBP to bind to DNA, and dimer dissociation is slow. This suggests that TBP dimerization may represent a mode of negative regulation of TBP activity.

    • Coleman RA, Pugh BF: Evidence for functional binding and stable sliding of the TATA binding protein on nonspecific DNA. J Biol Chem 1995, 270:13850-13859. TBP is shown to bind stably to non-specific DNA. Whether TBP is bound to TATA or to non-specific DNA, it is shown to be unusually stable with respect to dissociation, resistant to thermal denaturation, and capable of setting up functional pol II transcription complexes. These findings suggest that TBP binds to TATA and TATA-less promoters in the same way.

The X-ray crystal structure of the conserved carboxy-terminal core of human TFIIB is presented in complex with TBP and a TATA oligomer. The two imperfect amino acid sequence repeats of TFIIB form similar domains that straddle the carboxy-terminal, or upstream, stirrup of TBP. TFIIB also makes phosphate contacts with DNA both upstream and downstream of TBP.

    • Swaffield JC, Melcher K, Johnston SA: A highly conserved ATPase protein as a mediator between acidic activation domains and the TATA-binding protein. Nature 1995, 374:88-91. The yeast protein Sug1 is shown to interact with TBP in vivo and in vitro, and functions as a subunit of a transcriptional mediator by interacting with the activators Gal4 and VP16. Interestingly, Sug1 is an ATPase, which suggests that ATP hydrolysis might be involved in the activation process.

    • Connecting a promoter-bound protein to TBP bypasses the need for a transcriptional activation domain
    • •], demonstrates that tethering TBP to a sequence-specific DNA-binding domain obviates the need for an activation domain. The results suggest that recruitment of TBP is rate-limiting in vivo, and that the targeting of factors that function after TBP recruitment can be bypassed.

    • A general mechanism for transcriptional synergy by eukaryotic activators
    • Chi TH, Lieberman P, Ellwood K, Carey M: A general mechanism for transcriptional synergy by eukaryotic activators. Nature 1995, 377:254-257. This paper focuses on the mechanism of synergy by the activators VP16 and Zebra. When bound to multiple sites on the promoter, these activators enhance the formation of a TATA-TFIID-TFIIA complex. In addition, the activators enhance the recruitment of TFIIB into this complex. This paper demonstrates that activator synergism can arise both through interactions with a single target and through interactions with a series of targets.
