Nature Genet 1995, 11:462-464 Just when a pattern was beginning to emerge, with mutations of FGFR 1 and 2 in craniosynostosis and of FGFR3 in short-limbed dwarfism, this paper appeared, reminding us how little we really understand about molecular pathogenesis in the FGF/FGFR signalling pathway. A subset of Crouzon patients with the skin disorder acanthosis nigricans have an amino acid substitution just 11 residues away from that causing achondroplasia. This discovery broadens our perspective on FGFR3-related disorders and may be of particular value for investigating the mechanisms of acanthosis nigricans.

Nature Genet 1995, 9:321-328. Thanatophoric dysplasia is a sporadic, lethal bone dysplasia associated with very short limbs; cloverleaf skull is present in about 60% of cases. This elegant paper employed state-of-the art DNA sequencing techniques to identify FGFR3 mutations in both the extracellular and tyrosine kinase domains. The latter finding begs the question as to whether tyrosine kinase mutations of FGFR1 and FGFR2 await discovery.

Crouzon syndrome was associated with earlier sagittal suture closure (median 6 months) and a 73% prevalence of chronic tonsillar herniation; corresponding figures for Apert syndrome were 51 months and 2% respectively. Crouzon patients with tonsillar herniation had earlier closure of the lambdoid suture than those who did not.

This abstract describes the preliminary characterisation of the new mouse mutant Bulgy-eye (Bey), which has a retroviral vector inserted 6kb upstream of Fgf4, between the tandem Fgf3 and Fgf4 genes. It is suggested that FGF4 is most likely the ligand responsible for the Bey phenotype, but evidence for this has not yet appeared in print.

By five days, bony fusion across the coronal suture occurs in the absence of dura mater; in its presence (even when separated by a polycarbonate filter), patency is maintained.
