Transplantation 1995, 60:158-171. This interesting study compares the ability of different organs to induce tolerance to donor alloantigens after transplantation when survival of the primary grafts is induced by administration of a short course of an immunosuppressive drug, tacrolimus. The data presented show very clearly that all organs are not equivalent in their ability to induce tolerance. The authors try to assess how complete the degree of tolerance induced by the different strategies investigated is by using retransplantation.

The data support the view that there are multiple mechanisms by which tolerance to an allograft can be induced.

An examination of the mechanisms responsible for the induction of tolerance in vivo as a result of blocking the delivery of the second signal using CTLA4-Ig. The immunohistochemical data presented in the paper show that the leukocyte-infiltrating grafts in rats treated with CTLA4-Ig express Th2 cytokines. The authors suggest that deviation of the immune response to a Th2 cytokine phenotype is responsible for the induction of tolerance using this strategy.

This interesting study explores the use of gene therapy to deliver cytokine genes to tissue immediately before transplantation in an attempt to prevent graft rejection. The data presented show that expression of viral interleukin-10 and TGFβ in heart tissue before transplantation can lead to prolongation of graft survival.
