This paper compares the stability of isolated monomeric coat protein with the phage shells. While the coat protein was pressure denatured in a relatively low pressure range, the shells were pressure-resistant, only dissociating at subzero temperatures. Comparison of the final coat protein state obtained under pressure with the one obtained in the presence of chemical denaturant showed that the former was partially structured and that pressure probably denatures the coat protein by a domain mechanism.

Thermal denaturation resulted in a decrease in molar volume and an increase in adiabatic compressibility. In contrast, guanidine-hydrochloride denaturation caused a decrease in both molar volume and compressibility. The data suggest molten-globule intermediates.

Based on the argument that the values of heat absorbed from and released to the environment in a chemical reaction are not experimentally separable, it is proposed that entropy and enthalpy can only be determined if a molecular hypothesis relating them is made. A previously described model [22] is further developed in which enthalpy and entropy changes on reaction are determined by the thermally dependent probability of bond breakage. The most striking result, when the treatment is applied to data on association of several dimeric, trimeric and tetrameric proteins, is that the large entropy increase that drives protein association results from the conversion of protein-water bonds into the weaker protein-protein bonds.
