X-Gal (5-bromo-4-chloro-3-indolyl β-D-galactopyranoside) staining in embryos was performed as described in (8), but sometimes we used antibody to β-galactosidase (anti-β-Gal) and standard methods. For better staining of adult cuticle with X-Gal, pharate adults were extracted from the puparium and treated as described in (16).
