    • Fargeas M-J, Theodorou V, More J, Wal J-M, Fioramonti J, Bueno L: Boosted systemic immune and local responsiveness after intestinal inflammation in orally sensitized guinea pigs. Gastroenterology 1995, 109:53-62. This study shows that increased epithelial permeability, caused by TNBS inflammation in the proximal small intestine, results in a greater degree of sensitization in guinea pigs following drinking of cow's milk than in nontreated guinea pigs. Subsequent colonic antigen challenge results in greater ion secretion in those guinea pigs compared with the control group. Therefore, this study suggests that epithelial barrier defects may have significant consequences.

    • McKay DM, Croitoru K, Perdue MH: T cell-monocyte interactions regulate epithelial physiology in a co-culture model of inflammation. Am J Physiol 1996, 270:C418-C428. This study illustrates T-cell-to-monocyte communication in the induction of epithelial transport and permeability abnormalities. Experimental evidence is presented showing that specific T-cell activation leads to the release of IFN-γ, which activates monocytes, causing TNF-α production. This intercellular signaling results in epithelial pathophysiology in the absence of major cytotoxic effect.

    • Tolerance towards resident intestinal flora in mice is abrogated in experimental colitis and restored by treatment with interleukin-10 of antibodies to interleukin-12
    • Duchmann R, Schmitt E, Knolle P, Zumbuschenfelde KHM, Neurath M: Tolerance towards resident intestinal flora in mice is abrogated in experimental colitis and restored by treatment with interleukin-10 of antibodies to interleukin-12. Eur J Immunol 1996, 26:934-938. Intrarectal instillation of TNBS to Balb/c mice resulted in loss of local and systemic tolerance to bacterial sonicates. Concomitant treatment (intraperitoneal injection) with recombinant IL-10 or antibodies to IL-12 along with TNBS restored tolerance (splenocytes, lymphocytes from small and large intestine) to bacterial sonicates from autologous intestine.

This paper reports that severe combined immunodeficiency mice reconstituted with T cells and treated with IL-8 initially show neutrophil recruitment (within 4 hours) followed by a later (72-hour) accumulation of T cells at the site of cytokine administration.

Gastroenterology 1996, 110:1368-1378. This paper highlights the putative role of the intestinal epithelium in celiac disease, a condition generally considered a T-cell disorder. Data are presented showing that increased major histocompatibility class II expression on the epithelium precedes full T-cell activation in response to gliadin.

Gastroenterology 1995, 108:1688-1699. Compared with dexamethasone, an orally delivered dexamethasone-β-D-glucuronide conjugate is more potent in improving colonic water absorption and reducing the area of ulceration and histologic scope in rats treated with 4% acetic acid. The prodrug also maintains the levels of adrenocorticotropic hormone and corticosterone at close to normal.
