Demonstration of a phenotype-specific effect of tumor necrosis factor alpha on monocyte or macrophage apoE expression. Whereas tumor necrosis factor alpha could inhibit apoE expression in fully differentiated macrophages, it stimulated apoE expression in the monocytic or early macrophage phenotype. Such stimulation largely occurred at a level of apoE gene transcription.

Bone marrow transplanted from wild type mice to apoE null mice was demonstrated to reduce circulating cholesterol levels and reduce the extent of atherosclerosis. These benefits were achieved even though circulating apoE levels were only 10-15% of normal.

Transplantation of bone marrow from normal mice to apoE null mice reduced circulating lipid levels and protected against atherosclerosis. The results of this study and the one above demonstrate that macrophage-derived apoE can play a significant role in organismal lipoprotein transport and vessel wall homeostasis.

J Clin Invest 1995, 95:469-476. A transgenic mouse was developed that expressed high levels of apoE in the arterial wall while expression was low in other tissues. These mice were created on a background of normal hepatic apoE expression so they showed no difference in lipoprotein profile compared with wild type mice on normal and atherogenic diets. Overexpression of apoE in the vessel wall conferred significant resistance to the development of atherosclerosis and appeared to enhance efflux of cholesterol out of the vessel wall.

J Clin Invest 1995, 96:2170-2179. This study utilized apoE null mice and examined the effect of macrophage-specific apoE expression on vessel wall disease. Atherosclerotic lesion development was significantly less in mice expressing macrophage-specific apoE compared with littermates controlled for cholesterol level. These results convincingly demonstrate that macrophage-derived apoE is anti-atherogenic even in the presence of high levels of circulating atherogenic lipoproteins.

This study showed that apoE produced by macrophages is sequestered in a pericellular proteoglycan matrix. ApoE trapped in this matrix is subject to rapid reuptake and degradation by the macrophage. Alteration of composition of the pericellular matrix which accompanies macrophage differentiation or activation will therefore post-translationally modulate net secretion of apoE.
