This group compared the levels of oxidized lipids and antioxidants of normal and atheromatous human arteries, taking care to assess their recoveries during tissue processing. Atherosclerotic plaques were not deficient in the antioxidant vitamins C and E, despite the presence of large amounts of oxidized lipids.

A monoclonal antibody specific for hypochlorite-modified proteins stained atherosclerotic lesions more strongly than normal arteries, providing evidence for this oxidant in vivo. The staining was predominantly associated with cells (endothelial, macrophage and smooth muscle). Hemosiderin in macrophages was also observed.

Circulation 1995, 92:898-903. Rabbits fed 1% cholesterol for 1 month were returned to a normal diet for a further month. In this group, plasma cholesterol, elevated superoxide release and impaired response to endothelium-dependent vasodilators returned to normal. The major source of increased superoxide release in hypercholesterolemic vessels appeared to be the endothelium and not macrophages.

Hamsters on a moderately hypercholesterolemic diet deficient in antioxidants developed aortic lesions. These were prevented by α-tocopherol or probucol, whereas the same agents were not effective in more severe hypercholesterolemia. This suggests that hyperlipidemia may outweigh antioxidant effectiveness, which may explain some the variability seen in the efficacy of antioxidants as inhibition of atherosclerosis in experimental animal models.
