Mechanisms underlying the monocyte-mediated antibody-dependent killing of Plasmodium falciparum asexual blood stages

Studies of antibody-dependent cellular inhibition mediated by monocytes against P. falciparum in vitro suggesting that the merozoite, rather than the infected erythrocyte is the target of antibody-dependent cellular inhibition, and that antibody-dependent cellular inhibition is upregulated by IFN-γ and downregulated by IL-4.

Proc Natl Acad Sci U S A 1995, 92:7100-7104. The authors of this study describe genes expressing proteins resembling those involved in merozoite-erythrocyte binding. -
Baird JK, Basri H, Subianto B, Fryauff DJ, McElroy PD, Leksana B, Richie TL, Masbar S, Wignall FS, Hoffman SL: Treatment of chloroquine resistant Plasmodium vivax with chloroquine and primaquine or halofantrine. J Infect Dis 1995, 171:1678-1882. This study reports on the efficacious treatment of chloroquine-resistant P. vivax in northeastern Irian Jaya, Indonesia, using either of two regimens of chloroquine plus primaquine (including two doses once a day of 1 mg/kg plus 0.5/mg/kg primaquine administered in conjunction with chloroquine), or standard halofantrine therapy.

Atovaquone and proguanil for Plasmodium falciparum malaria
-
Radloff PD, Philipps J, Nkeyi M, Hutchinson D, Kremsner PG: Atovaquone and proguanil for Plasmodium falciparum malaria. Lancet 1996, 347:1511-1514. A trial of atovaquone plus proguanil among 71 Gabonese adults showing an 87% cure. However, only one of 71 had a recurrent parasitemia and this could have been a reinfection. The remaining subjects 'not cured' were actually lost to follow-up. Thus, 62 of 63 subjects (98%) enrolled and who completed the study did not have recurrent parasitemias within 28 days of therapy.

Inhibition of Plasmodium falciparum malaria using antisense oligonucleotides
-
Barker RH Jr, Metelev V, Rapaport E, Zamecnik P: Inhibition of Plasmodium falciparum malaria using antisense oligonucleotides. Proc Natl Acad Sci U S A 1996, 93:514-518. Without phosporothioation of antisense oligodeoxynucleotides (PS AS-ODNs), inhibition of P. falciparum in-vitro was not sequence-specific. With phosphorothioation of AS-ODNs, inhibition occurred at lower concentrations and in a sequence-specific manner.
