Coronal brain sections (10 or 30 μm thick) were obtained with the use of a cryostat (0.5 to 1 mm anterior to bregma for striatum; 3.8 to 4.3 mm posterior to bregma for other brain regions including hippocampus) and mounted on glass slides. Without fixation, tissue sections were immersed in a solution of TSQ (4.5 μm, Molecular Probes) in 140 mM sodium barbital and 140 mM sodium acetate buffer (pH 10) for 90 s. After washout with normal saline, TSQ fluorescence was observed under fluorescence microscopy with an ultraviolet filter (excitation 355 to 375 nm; dichroic, 380 nm; barrier, 420 nm).

    • 2+ chelation on transient ischemia, 5μl of normal saline (control) or 100 mM CaEDTA or ZnEDTA in saline was injected into the lateral ventricles of rats 30 min before the beginning of ischemic insults. The procedures were conducted by an investigator blind to drug identity. Selection of anesthesia (pentobarbital versus isoflurane) had no gross effect on the translocation of TSQ fluorescence or CaEDTA neuroprotection reported here. CaEDTA injection did not alter temporalis muscle temperature.
