31. Selective homogenization of stem tissue is sometimes desirable to avoid releasing compounds located within the wood. The milling of stems with sufficient force to homogenize only the bark tissue alleviates the need for preparative bark stripping.
32. Milling of 15- to 20-g portions gives optimum homogenization in a mill of this size.
33. It may be necessary to periodically cool the chamber and piston between loads to maintain an appropriate temperature.
34. It is worth noting at this point that the limiting factor in microsome preparation at this scale is usually not the homogenization method but the time and capacity needed for centrifugation.

36. The rapid addition of large amounts of liquid nitrogen in order to speed up the cooling process may result in damage to the mill.
37. For large preparations, it may be necessary to periodically empty the collection box as the grinding chamber will become clogged if the collection box is overfilled.
