Tissue Antigens 1995, 45:169-176. Subtypes of B27 were investigated in two groups of healthy (n = 17) and AS patients (n = 45) from Asian Indian and Thai populations. B*2706 was found in 47% of the healthy Thai individuals and was absent in patients. The negative association described suggests a protective effect of this subtype on AS susceptibility in Thai patients. B*2707 was found to be associated with AS in the populations studied.

Immunogenetics 1995, 42:123-128. Binding of polyalanine-derived nonapeptides carrying Arg at P2 and a different amino acid at P9 by B*2705 and B*2702 were analyzed. Peptides with basic residues, aliphatic and aromatic at P9, have similar affinity in the binding of B*2705. However, B*2702 binds well to P9 aliphatic and aromatic peptides but only very weakly to basic peptides. Substitution at position 81 can explain the differences for both B*2702- and B*2705-bound peptides.

A molecular dynamic simulation method based on the MHC-ligand complex was used to predict the role of secondary anchor residues for binding to B27. Several nonnatural peptides were designed to optimize the P3 interactions with pocket D, showing that a bulky hydrophobic side chain at P3 enhances the binding to HLA-B27, Furthermore, the putative TCR contact (P4-P8) was replaced with organic fragment without altering the affinity of this designed ligand. The combination of these strategies opens new possibilities for designing peptides for use in immunotherapy.

Eur J Immunol 1995, 25:2370-2377. The contribution of major anchor residues 2 and 9, the role of pocket B structure, and the binding properties of B*2703 were analyzed. Synthetic peptides presented by B*2705 demonstrated that Arg2 is not absolutely required because Gln2 analogues also bind. Changes affecting positions 2 and 9 suggested that B*2703 is probably the most stringent specificity for the Arg2 peptide motif. The binding dependence of the B*2703 subtype can be explained by the molecular effect of the His59 change.

Coexpression in NW lymphoblastoid cells of B8 and B*2702 prevented the presentation on HLA-B8 influenza A nucleoprotein-restricted epitope by HLA-B8, although added peptide could still be presented. The results suggest competition mechanisms occurring between the two class I molecules in the endoplasmic reticulum.

Eur J Immunol 1995, 25:2479-2485. An analysis of TCRs in HLA-B27 (B*2705) alloreactivity was carried out in CTLs from two individuals. Both α and β chain usage was nonrandom, but there were significant differences. Preferential Vα usage was associated with particular individuals. In contrast, preferred usage of Vβ subgroup 4 was a major donor- and epitope-independent feature, suggesting an involvement in the recognition of the HLA-B27 molecule.

Eur J Immunol 1995, 25:3199-3201. Subtypes of HLA-B27 were analyzed in groups of 40 healthy individuals and 35 Sardinian AS patients. B*2709 accounts for 25% of the HLA-B27 subtypes in this population. None of the patients were found to be positive. B*2709 differs from the widespread AS-associated B*2705 allele for a single substitution (His vs Asp) that can be relevant in AS susceptibility.

Lancet 1995, 345:1542-1544. Several short peptide sequences are shared between the first hypervariable region of B27 and proteins of gram-negative bacteria. The B*2705 sequence contains a nonapeptide, LRRYLENGK, predicted to bind in the binding cleft of B27. Some nonapeptides from enteric bacteria that share sequence with B27 were tested for binding by B27 with an assembly assay. Thus peptides that both mimic and bind B27 may constitute the molecular components of the mechanism of spondyloarthropathies.

FEBS Lett 1995, 369:243-248. In this study a computer search has revealed a novel homology between pulD K. pneumoniae secretion protein and B*2705. Additionally, molecular mimicry has been described between pullulanase enzyme (pul A) and types I, II, and III collagen. Furthermore, the level of cross-reactive antibodies found in AS patients could be relevant in the development of the disease.

The frequency of ankylosing enthesopathy is dependent on the H-2 type and the presence of HLA-B27. Its relation to age, gender, environment, and immunology is considered.
