Using extraction with mild detergents and fluorescence recovery after photobleaching, they show that phosphorylation attaches AChRs in the cytoskeleton. The authors propose that localized activation of the agrin receptor leads to a localized tyrosine phosphorylation and attachment of AChRs.

This paper describes tissue culture experiments showing that agrin inhibits neurite outgrowth of chick ciliary ganglia neurons and induces the formation of presynaptic specializations. Unlike AChR-aggregating activity, agrin's effect on the presynaptic cell is not influenced by splicing at sites A and B (see Figure 1). These results are the first evidence that agrin may play a role in the formation of both post- and presynaptic specializations.

In these mice, no AChR aggregates are formed, indicating that rapsyn plays a fundamental role in the formation of AChR aggregates. Synapse-specific gene transcription of AChR subunits is, however, unaffected. Moreover, the extent of nerve branching is increased, suggesting that a retrograde signal is involved in presynaptic differentiation.

The authors show that neurite outgrowth of motor neurons on several, but not all, ECM substrates is inhibited by a recombinant carboxy-terminal fragment of laminin β2 (see Figure 3). Inhibition of neurite outgrowth is selective for motor-neuron-like cells and is mediated by the tripeptide LRE. The authors propose that LRE may be a muscle-derived signal that plays a role in the formation of presynaptic specialization in the nerve terminal.
