On the basis of the presence of docked vesicles in the absence of VAMP or syntaxin, it has been stated that SNARE proteins are not involved in docking (39), despite the proven need for SNAREs in docking both ER- and Golgi-derived vesicles (23, 25). There are many possible explanations. The most likely is that docking is mediated by binding of synaptotagmin to SNAP-25, which occurs with high affinity in vitro (G. Schiavo and J. E. Rothman, unpublished data). Another, nonexclusive, possibility is that synapses possess an additional specialized docking mechanism that acts before SNAREs, indeed, synaptic vesicles dock only at certain locations, even though syntaxin and SNAP-25 are present along the axon. The most trivial explanation would be that additional VAMP or syntaxin homologs replace those deleted or cleaved away.

Evidence originally interpreted as establishing bulk flow as the major mechanism of transport from ER to Golgi (46) in fact favors transport mediated by transport signals, when reinterpreted in light of more recent data. Tripeptides with the minimal structure for N-glycosylation enter cells and become N-glycosylated in vivo in the ER. Glycotripeptide secreton was shown to be as rapid as secretion of the fastest secreted proteins, from which we concluded that such proteins must exit the ER in vesicles at their prevailing concentration [for example, by bulk flow (Fig. 3C)]. Although these measurements remain valid, our conclusion was in error because it did not take into account the time now known to be required for protein folding after translocation. For example, viral glycoproteins require ∼5 min to fold but are then exported within 1 min after folding [A. Helenius, Mol. Biol. Cell 5, 253 (1994)]. Our original bulk flow transit time (5 min) together with a residence time of 1 min after folding in the ER imply that these proteins are exported at 5 to 10 times their bulk concentration in ER.
