Alanine mutants of ANP were displayed on the pB1537 phagemid that contained two additional mutations (R3D in ANP and Q29E in the spacer residue immediately after residue 28 of ANP) that increase the efficiency of ANP display without affecting NPR-A-binding affinity (B. Li et al., unpublished data). For Fig. 2 (upper panel) affinities were based on phage ELISA measurements, except those for F8A and I15A, which were determined from radioimmunoassay (RIA) measurements of the respective peptides.

Peptides were synthesized on an Applied Biosystems 430A automated synthesizer according to standard N-tert-butoxycarbonyl chemistry protocols, cleaved off the resin with anhydrous HF at 0°C, then cyclized by the potassium ferricyanide method and purified by reversed-phase high-performance liquid chromatography. Masses of each peptide were verified by mass spectrometry. All the peptides except for variant 6 were synthesized with an amidated COOH-terminus.
