    • Comparative chromosome painting discloses homologous segments in distantly related mammals
    • To achieve significant painting on non-primate chromosomes with human probes the original protocols were changed to allow stringent hybridization conditions. Most efficiently, increasing human DNA probe concentration or a prolonged hybridization time gave FISH signals strong enough to analyze chromosome rearrangements.

    • Visualisation of the conservation of synteny between humans and pigs by heterologous chromosomal painting
    • This is the first complete comparison of the human and a non-primate karyotype using chromosome painting. However, as the authors state, some of the human paints gave only poor hybridization signals on homologous pig chromosomes. As many of these libraries were made from human-rodent hybrid cell lines there is a significant amount of contamination of non-human DNA in the paints which may alter the specificity of the probes.

    • The origin of human chromosome 2 analyzed by comparative chromosome mapping with a DNA microlibrary
    • Chromosome banding has shown a small inversion in the pericentric region of the primate chromosome 2p homologs of the chimpanzee which could be interpreted as a common derived trait of human and chimps. However, chromosome painting, with a microdissection library from the entire long arm of human chromosomes, to ape and monkey chromosomes showed that this inversion evolved independently in different phylogenetic lines.

    • Subregional or band specific paints can be obtained from human-rodent somatic cell hybrids because human chromosomes are often rearranged in these cell lines. To produce painting probes, Alu-PCR allows the DNA of the human chromosome fragments to be extracted from the rodent background.

    • A comprehensive human linkage map with centimorgan density
    • This article, based on the work of more than 100 collaborators presents recent advances in the large scale mapping of the human genome. For comparative purposes, human polymorphic markers which are the main surce of the human high density genetic map may only be useful within higher primates because they are not sufficiently conserved to recognise locus homology in more distant related species. However, YAC clones containing Généthon-based STRP markers can be used for subregional chromosome painting in more distantly related mammals. These data could be used to compare the human genetic and chromosome map with the chromosomes of non primate mammals.
