Clathrin-independent pinocytosis is induced in cells overexpressing a temperature-sensitive mutant of dynamin
-
of outstanding interest, The kinetics of horseradish peroxidase (HRP) uptake were examined in cells expressing a temperature-sensitive mutant of dynamin that display a rapid and reversible block in clathrin-mediated endocytosis. At early times after shift to the non-permissive temperature, HRP uptake was partially inhibited but recovered to wild-type levels within 30 min, even though clathrin-mediated endocytosis remained blocked. The results suggest that a clathrin-independent pinocytic pathway is induced to compensate for the loss of clathrin-dependent endocytosis.

of special interest, Using a cell-free assay, we demonstrate that kinase-deficient epidermal growth factor receptors (EGF-Rs) are unable to undergo ligand-induced recruitment into coated pits. Addition of a soluble EGF-R kinase domain rescues this deficiency in trans. Evidence that tyrosine kinase substrate(s) other than the EGF-R itself are required for efficient endocytosis is presented.
