GAP-43 transgenic mice: dispersed genomic sequences confer a GAP-43-like expression pattern during development and regeneration
-
of special interest, This paper identifies regulatory sequences that can direct GAP-43 transgene expression in a manner that closely mimics the endogenous gene, and shows that a neuron-restrictive regulatory region in the first intron is necessary to prevent non-neuronal expression of GAP-43.

Cell-specific expression from the human dopamine β-hydroxylase promoter in transgenic mice is controlled via a combination of positive and negative regulatory elements
-
of special interest, The authors provide evidence for negative regulation among neuronal subtypes that restricts DBH expression to catecholaminergic neurons. The paper also illustrates the value of β-galactosidase as a reporter for determining which neuronal populations express a transgene.

The neuron-restrictive silencer factor (NRSF): a coordinate repressor of multiple neuron-specific genes
-
of outstanding interest, The isolation of cDNAs encoding human NRSF, a silencer factor for the SCG10 gene. NRSF was found to bind potential NRSEs in three other neuronal genes, and its mRNA was found to be expressed in many non-neuronal tissues and in neuronal precursors, but not in neurons. The NRSF gene is identical to the REST gene that was isolated independently by Chong et al. [30].

REST: a mammalian silencer protein that restricts sodium channel expression to neurons
-
of outstanding interest, The isolation of the human REST gene as a silencer-binding protein for the NaII gene. A truncated version of REST was shown to lack repressor activity and antagonize the endogenous factor. At embryonic day 11.5 of the mouse, REST mRNA was shown to be ubiquitously expressed except in differentiated neurons. The REST gene is the same as the NRSF gene independently isolated by Schoenherr and Anderson [29].

NF1-L Is the DNA-binding component of the protein complex at the peripherin negative regulatory element
-
of outstanding interest, The identification of a component of a repressor complex that negatively regulates the peripherin gene. This component, NF1-L, a member of the CTF/NF-1 transcription factor family, becomes dissociated from the repressor complex during induction of peripherin by NGF in PC12 cells, thereby allowing full activation of the gene.

Cell-specific action and mutable structure of a transcription factor effector domain
-
of special interest, This paper demonstrates that transcriptional activation by SCIP is dependent upon an amino-terminal SCIP effector domain and does not require the POU domain. This same SCIP domain mediates transcriptional repression, which is thought to occur by a ‘quenching’ mechanism that is dependent on promoter context. Transactivation by the SCIP domain is also shown to be cell type specific.
